ACCUTASE™ 细 胞 消 化 液

— 传统贴壁细胞消化（胰酶/EDTA）的完美替换产品

描 述：

Mpbio公司提供的ACCUTASE™具有蛋白酶和胶原酶活性，是胰酶/EDTA的替换产品，用于从常规组织培养器皿和粘附培养器皿中消化细胞。另外进行细胞计数、转染或分析检测（如流式细胞术）前，常加入一定量的accutase，将细胞集落 (clumps)分散成单个细胞然后进行下游实验。Accutase因其消化方式温和，对细胞伤害极低，不含任何动物或细菌来源组分等特点成功替代传统消化产品，广泛应用于常规贴壁细胞（血清培养细胞和无血清培养细胞）的消化，以及成为干细胞（如hESCs、mESCs等）传代培养最受欢迎的产品之一。即用型的无菌产品，使用非常方便。

配 方：

Accutase具蛋白酶和胶原酶活性，以Dulbecco’s PBS溶液（含0.5 mM EDTA·4Na和酚红）形式供应。

优 势：

● 应用于绝大多数原代细胞和哺乳动物细胞系的消化

实验验证的细胞系（cell line）：成纤维细胞,角蛋白细胞,血管内皮细胞,肝细胞,血管平滑肌细胞,肝细胞祖细胞,原代鸡胚神经细胞,骨髓干细胞,贴壁CHO和BHK细胞,巨噬细,293细胞,L929 细胞,永久性小鼠睾丸生殖细胞，3T3,Vero ,COS, HeLa,NT2,MG63, M24,A375转移性黑色素瘤,神经胶质瘤U251和D54,HT1080纤维肉瘤细胞,Sf9 昆虫细胞。（参考文献1-9）

● 温和有效的消化胚胎干细胞（hESCs和mESCs）,冻存后细胞具更高复苏率（图2，文献10）

● 几分钟内实现粘附细胞的分离，不需清洗或中和反应，节省细胞传代时间

● 温和而快速的分离贴壁细胞，不会破坏流式细胞术检测的表面抗原

● 温和消化维持最高细胞活力，即使长时间消化（45 min）细胞活力达97 ± 3%（图1）

● 同时结合胶原酶和蛋白酶活性，不含哺乳动物、细菌来源产品或昆虫杆状病毒表达系统组分

应 用：

（1）替换胰酶/EDTA（Replacing Trypsin/EDTA）

Accutase的用量和方法不需做改动，特别适用于在无血清和无蛋白培养基等不含血清的培养基内贴壁细胞的分离。相比于胰酶/EDTA，优点在于：即用型，使用方便；大大降低对细胞的伤害；提高细胞产量及细胞活力；增高铺板效率；改善细胞形态（定性）和细胞生长特性。

（2）功能分析（functional assays）

细胞表面标志物分析，流式细胞术，血清饥饿法检测细胞静止状态，细胞增殖，转染实验，细胞趋触分析，细胞、神经嵴迁移实验，肿瘤细胞迁移。

（3）组织解离（tissue disaggregation）

在4-25℃下进行组织解离，因酶的特性切忌在37℃操作。根据组织类型和解离温度优化解离时间。一般情况下，4℃过夜孵育（12-16 hrs），25℃孵育1-2 hrs。

应用实例：

操作步骤：（快速简单）

（1）37℃或室温溶解Accutase，用无菌PBS溶液清洗细胞培养容器（培养板、培养瓶等） 。

（2）以无菌操作的方式按 75 cm²表面积加入10 ml Accutase的量覆盖贴壁细胞；重新放入37℃培养箱内，消化15-20 min。（消化时间可自行调整。大部分细胞处于Accutase中高达1小时，对细胞无影响）

（3）消化结束后，依常规操作进行细胞计数和传代：不需另做清洗和酶活终止步骤。
